The adult hippocampus is one of the primary neural structures involved in memory formation. Within the hippocampus, the DGdentate gyrushas the potential to make a preferential contribution to neural circuit modification. To better understand how the DG regulates hippocampal activity, we investigated the reciprocal projection of bilateral DG and the microcircuits between various types of cells in the DG.

First, we used immunofluorescence method to study the composition of neurons in DG, and found that there are mainly three kinds of mature neurons in DG, namely SST, CR and CB. CB neurons are distributed in GrDG, SST neurons and CR neurons in poDG. To further investigate the projection relationship between these types of neurons. Anterograde AAV-DIO-EGFP was injected into the hippocampal DG of transgenic mice whose CR neurons specifically expressed CRE. We found that the fibers of CR neurons can reach the outer edge of ipsilateral and contralateral GrDG. Trans-synaptic RV was used to demonstrate the presence of neuron-to-neuron projection of bilateral poDG. Finally, we used transgenic mice combined with trans-synaptic RV and immunohistochemical method to prove the existence of CR to SST, CB to SST projection microcircuits in DG. In conclusion, we use the neural tracer method to more clearly clarify the bilateral projection and the projection microcircuits among various types of neurons in hippocampal DG.
