Abstract:
Natural Products kept promising compound libraries for searching new anti-cancer agents. In this study, about 40 compounds were screened for their potential anti-tumor activities, and S5 (chalcomoracin, isolated from mulberry) showed strong cytotoxities (IC50mM) on several cell lines in vitro, including MDA-MB-231, PC-3, and HCT-116 tumor cells. Moreover, it induced cell death by characteristic paraptosis pathway. Molecular mechanisms of S5 were further investigated, and PINK-1 was proved to induce mitophagy , which influenced sequential downstream signals including Alix, GRP78, Chop etc., and resulted in endoplasmic reticulum stress, multivesicular bodies and final paraptosis and cell death. Detailed molecular pathway of paraptosis indused by S5 are now under further investigation. 5-8
